GFP-FRET detects swings of the lever-arm of myosin motor

摘要
The "lever-arm" model of a myosin motor predicts that the lever-arm domain in the myosin head tilts and swings against the catalytic domain during ATP hydrolysis, resulting in force generation. This paper describes the experiments of the fluorescence resonance energy transfer (FRET) between two fluorescent proteins (GFP and BFP) fused to the N- and C-termini of the Dictyostelium myosin motor domain to detect such swings of the lever-arm during the ATP hydrolysis cycle.
