Ligninolytic Properties of Highly Expressed Transformant with Novel Lignin Peroxidase from Hyper Lignin-Degrading Fungus Phanerochaete sordida YK-624

摘要
The ligninolytic properties of A-11, a YK-LiP2 over-expressed transformant from hyper lignin-degrading fungus Phanerochaete sordida YK-624, were determined. This transformant showed higher ligninolytic activity by 7.6% and selectively than the wild type of P. sordida YK-624 in beech wood meal after 4-week incubation. LiP activity in A-11 cultured in the wood meal was also higher than that of the wild type, and the constant expression of recombinant ylpA was demonstrated by transcriptional analysis. Moreover, high glucose conversion was observed in the enzymatic saccharification of the wood meal which was treated with A-11, compared with that of the wild type-treated wood meal. These results suggest that high expression of YK-LiP gene in P. sordida YK-624 improves both ligninolytic ability and saccharification of the fungal treated-wood meal.
