By introducing instrumental characteristic factor and molecular absorption correction factor, herein we discuss the effects of testing sensitivity of spectrofluorometer and molecular absorption on the resonance light scattering spectra ( RIS) of rhodamine 6G as an example. A correction equation containing the two factors was proposed. It has been known that the molecular absorption results in the decrease of the RLS intensity in the wavelength region near absorption band, and leads to the pattern change of RLS spectra. By using the correction equation, both the instrumental and molecular absorption effects can be corrected, and the detection sensitivity can be improved.
