Identification of Sterol-Containing Domains in Vacuolar Membranes by means of Confocal Microscopy

摘要
The method of confocal microscopy and membrane probe Filipin were applied for the identification of sterol-containing domains in the tonoplast of beet root cells. The presence of sterol-containing domains in the tonoplast was confirmed using methyl-beta-cyclodextrin. It was found that sterol-containing vacuolar membrane included separate small sterol-enriched sites with a more intensive fluorescence; these sites can be attributed to raft structures. Our data demonstrate that the sterol content in isolated rafts can be assessed by confocal microscopy. A detailed analysis of the isolated rafts indicated that they contain (beta-sitosterol, stigmasterol, alpha-tocopherol, and cholesterol.
