Comparison of the determination of affinity constants with surface plasmon resonance and quartz crystal microbalance

A comparison between quartz crystal microbalance (QCM) and surface plasmon resonance (SPR) as devices for the determination of affinity constants is presented. The association and dissociation rate constants k(d) and k(a) respectively were determined by non-linear curve fitting of the time dependent measurement signals during association and dissociation processes. We used the virus-protein vp73 from the african swine fever (ASF) virus as analyte and immobilised an anti-ASF-antibody. The SPR measurements yielded 7.9.10(-4) s(-1) for k(d) and 5.6.10(3) (Ms)(-1) for k(a). The QCM results were 3.1.10(-4) s(-1) for k(d) and 1.1.10(4) (Ms)(-1) for k(a). k(d) and k(a) differ for about a factor of 2 for the QCM and SPR respectively.
