摘要
To find out the effect of vitrification on in vitro matured buffalo (Bubalus bubalis) oocytes, quality oocytes were collected from slaughterhouse ovaries and matured using the standard IVMFC protocol of our laboratory. Matured oocytes were vitrified by using 20% ethylene glycol + 20% dimethyl sulfoxide (DMSO) + 0.6M sucrose for a period of a minimum of 7 days in liquid nitrogen (LN2) at - 196 degrees C. Post thaw recovery of oocytes was 81.35%, and 18.60% of oocytes were lost during recovery. Major cryo injuries noted for these oocytes were zona breakage, crack in zona, leakage of the cellular contents and cytoplasmic shrinkage.
