Enzyme-linked Immunosorbent Assay for the Determination of Paeoniflorin in Plasma

A method was developed for the determination of paeoniflorin in plasma by competitive enzyme-linked immunosorbent assay(ELISA). The artifical antigen was used as immunogen and applied in New Zealand rabbits to get the anti-paeoniflorin antibodies. Hapten coupled with OVA was used as coating antigen. The linear concentrations ranged from 10 mu g/L to 200 mu g/L and the calibration curve was Y = -0.1721 logC +1.9645. Recoveries for the standard addition paeoniflorin to blank palsma were 89%-102%. The method has been successfully applied to determine the concentration of paeoniflorin in rat plasma. After an oral administration of the Si-Ni-San decoction, the bioavailability of paeoniflorin was lower than a single dose of paeoniflorin.
