Research of transformation efficients of Cm-AAT genes (1, 3 and 4) responsible for aroma synthesis in melon (Cucumis melo L., var. cantalupensis Naud)

摘要
In the research, Cm-AAT antisense genes responsible for synthesis of volatiles in melon (Cucumis melo L., var. cantalupensis Naud) were transformed by Agrobacterium tumefaciens. Commercial recombinant RNAi - pFGC5941 plasmid transformed from Escherichia coli (DH5 alpha) to Agrobacterium tumefaciens, was used. Based on the results, from the 1200 seeds used in the experiment, 58 putative transgenic plants for AAT1 gene, 39 putative transgenic plants for AAT3, 28 putative transgenic plants for AAT4 were obtained. These results were confirmed by PCR analysis. Transformation efficiency of antisense genes was found as 2.5% for AAT1, 1.7% for AAT3 and 1.25% for AAT4. Cotylesione and proximal explants from the seeds were cultured on the selective medium. Proximal explants gave better results in view of regeneration and shoot rates for all genes.
