Coumarin Derivatives from Hydrangea macrophylla and Evaluation of Their Cytotoxic Activity

摘要
The aim of the research is to evaluate the chemical constituents of potential extract of Hydrangea macrophylla. The cytotoxic screening indicated that methanol extract from the leaves of H. macrophylla exhibited the strongest cytotoxic activity against MCF-7 cells (IC50 value of 150.63 mu g/mL). Methanol extract of H. macrophylla was prepared by solid liquid extraction to give the crude extract. Isolation of secondary metabolites from the crude extract of H. macrophylla leaves using several chromatography methods including vacuum liquid and column chromatography yielded two phenolic compounds. Their chemical structures were identified based on basis of spectroscopic data interpretation evidence as umbeliferon (1) and thunberginol A (2). The cytotoxic activity of the compounds 1 and 2 from H. macrophylla was performed by the colorimetric method against MCF-7 breast cancer lines. The IC50 values of compounds 1 and 2 on MCF-7 breast cancer lines were 631.04 mu g/ml and 180.33 mu g/ml, respectively. Compound 2 showed stronger cytotoxicity than compound 1. To the best of our information, cytotoxic activity of the crude extract and isolated compounds from H. macrophylla against MCF-7 breast cancer lines is firstly reported in this study.
