Nanoparticles are rapidly adopted in pharmaceutical fields to develop drugs to control various diseases. Meanwhile, phyto-medicine also gains popularity due their potential pharmacological significance. Therefore, in present study, green synthesis of silver nanoparticles from aqueous extract of Cyathocline purpurea (CpAgNPs) was achieved and their efficacy against the MCF-7 breast cancer cell line was tested. The study revealed green synthesized silver nanoparticles were spherical in shape, crystalline in nature, and average size is 59.9 nm. At the 125 & mu;g/mL CpAgNP concentration maximum decrease in the percentages of viable cancer cells (0.5 & PLUSMN; 0.02%) was reported by MTT cytotoxicity assay. The Annexin-V flow cytometry (apoptosis assay) reported increased in early apoptotic cell by 8.7 & PLUSMN; 0.11% and late apoptotic by 19.6 & PLUSMN; 0.3% at IC50 concentration (67.31 & mu;g/mL), while Caspases-3 expression flow cytometry assay revealed increase in the percentage of active caspase-3-positive cells by 20.7% in CpAgNP-treated cells. Further, cell senescence assay also revealed that as compare to untreated (22 & PLUSMN; 0.58) cell CpAgNP-treated cells were shows increase (30 & PLUSMN; 0.58) in senescent cell. However, green produced nanoparticle-promoted cell death in MCF-7 breast cancer cells suggests that Cyathocline purpurea-mediated silver nanoparticles have anticancer potential. Therefore, the green approach of silver nanoparticle manufacturing was environmentally safe, nontoxic, affordable, and simple to use. These silver nanoparticles could be employed to treat breast cancer cells.
