Effects of methylprednisolone on apoptosis and expression of interleukin-1 β-converting enzyme in the kidneys of MRL-lpr/lpr mice

Objective To investigate the effects of methylprednisolone (MPS) on apoptesis and expression of interleukin-1 β-converting enzyme (ICE) in the kidneys of MRL-lpr/lpr mice suffering from lupus nephritis. Method Image analysis system, RT-PCR and Northern blot analysis were used. Results The number of apoptotic cells in glomerular mesangium increased significantly in the MPS group compared with those in the control group (P<0.05). The numbers of apoptotic cells of both the peri-tubular cells and peri-vascular infiltrating cells were also higher in the MPS group than in the control group (both P<0.01). Expression of ICE mRNA was higher in the treated than that in the control group. Conclusion These results suggest that MPS may induce both apoptosis and ICE mRNA expression in kidneys of MRL-lpr/lpr mice with lupus nephritis, implicating that apoptosis may be involved in the mechanism of MPS therapy for lupus nephritis.
