干扰NETO2通过调控JAK2/STAT3信号通路抑制肺腺癌细胞增殖和转移能力

摘要
目的 研究神经纤毛及唾样蛋白2(NETO2)在肺腺癌中的表达及临床意义,并探讨NETO2对肺腺癌细胞增殖和转移能力的作用及其机制。方法 采用免疫组化检测NETO2在肺腺癌组织中的表达水平,并分析NETO2的表达与肺腺癌患者临床病理参数及预后的关系。培养肺腺癌细胞A549,随机分为对照组(si-NC组)和实验组(si-NETO2-1组、si-NETO2-2组),分别转染NC siRNA和NETO2 siRNA-1、NETO2 siRNA-2,Western blot检测各组细胞中NETO2蛋白的表达水平;MTS实验和平板克隆实验检测各组细胞的增殖能力;Transwell实验检测各组细胞的转移能力;Western blot检测各组细胞中JAK2/STAT3信号通路蛋白JAK2、pJAK2、STAT3和pSTAT3蛋白表达。结果 免疫组化结果显示,与癌旁组织相比,NETO2在肺腺癌组织中的表达增加(P<0.05),NETO2高表达与肺腺癌患者肿瘤T分期、淋巴结转移和TNM分期均相关(P<0.05),与NETO2低表达的肺腺癌患者相比,NETO2高表达的肺腺癌患者生存预后较差;与si-NC组相比,si-NETO2-1、si-NETO2-2组A549细胞中NETO2蛋白表达减少(P<0.05);与si-NC组相比,si-NETO2-1、si-NETO2-2组A549细胞的增殖和转移能力降低(P<0.05),A549组细胞中JAK2/STAT3信号通路相关蛋白pJAK2和pSTAT3蛋白表达降低(P<0.05),JAK2和STAT3蛋白表达无明显变化(P>0.05)。结论 NETO2可能通过激活JAK2/STAT3信号通路促进肺腺癌的增殖和转移能力。
