Mesophyll protoplasts of two cultivars of Nicotiana rustica L. were cultured in Nagata-Takebe me-dium. The callus formed was inoculated onto a modified MS media （MSBI, MSBN） to induce organo-genesis. Greenish outgrowths, which developed from the callus of only one cultivar, were transferredagain to basic medium without plant growth regulators （MS）. These greenish spots developed further intoflower buds under various light conditions. Especially leafless flower buds formed directly from a partof greenish outgrowths on callus. They bloomed and set seeds within the test tubes.
