参薯是热带和亚热带的重要薯类作物，其块茎中富含淀粉。由于miR172是调控马铃薯块茎形成的关键基因，为分析miR172及其靶基因AP2的基因特征和不同发育时期的表达模式，本研究开展参薯miR172及靶基因预测分析和验证。基于参薯转录组和基因组序列，找到8个DalmiR172基因，并预测到其靶基因主要为AP2基因家族。通过多序列比对，发现8条DalmiR172成熟序列高度保守，其前体序列有一定差异。通过RT-qPCR分析参薯miR172在不同生长阶段中的表达情况，发现参薯已结薯植株中的表达量显著高于参薯幼苗期，表明miR172的表达水平在参薯幼苗到结薯时期呈增加趋势，推测miR172可能对参薯块茎形成具有促进作用。通过农杆菌介导的烟草瞬时表达实验验证了参薯miR172负调控AP2的转录水平。本研究为miR172-AP2调控模块在参薯生长发育过程中的功能分析提供实验依据。
