CircZNF609调控miR-22-3p对幼龄癫痫大鼠脑组织神经细胞凋亡及炎症反应的影响

摘要
目的 探讨CircZNF609调控miR-22-3p对幼龄癫痫大鼠脑组织神经细胞凋亡及炎症反应的影响。方法 取幼龄大鼠建立癫痫模型,随机分为模型组、CircZNF609敲低组、阴性对照组、CircZNF609敲低+miR-22-3pinhibitor组,每组12只,另取12只幼龄大鼠作为对照组,分组干预处理后,观察各组大鼠癫痫发作次数和平均发作时间;Morris水迷宫实验检测各组大鼠认知功能;尼氏染色观察各组大鼠海马神经元病理形态变化;TUNEL染色检测各组大鼠海马神经元凋亡;酶联免疫吸附法(ELISA)检测各组大鼠血清炎症因子前列腺素E2(PGE2)、白细胞介素(IL)-6、IL-17水平;实时荧光定量PCR法检测各组大鼠脑组织CircZNF609和miR-22-3p表达。结果 与对照组相比,模型组大鼠神经元严重损伤,癫痫发作次数与平均发作时间增多,逃避潜伏期延长,游泳时间百分比、海马神经元凋亡率、血清炎症因子PGE2、IL-6与IL-17水平、脑组织CircZNF609表达升高,脑组织miR-22-3p表达降低(均P<0.05)。与模型组相比,CircZNF609敲低组大鼠神经元损伤减轻,癫痫发作次数与平均发作时间减少,逃避潜伏期缩短,游泳时间百分比、海马神经元凋亡率、血清炎症因子PGE2、IL-6与IL-17水平、脑组织CircZNF609表达降低,脑组织miR-22-3p表达升高(均P<0.05);阴性对照组大鼠各指标无明显变化(P>0.05)。下调miR-22-3p表达可明显减弱敲低CircZNF609对癫痫大鼠脑组织神经细胞凋亡及炎症反应的抑制作用。结论 敲低CircZNF609可抑制幼龄癫痫大鼠脑组织神经细胞凋亡及炎症反应,减轻癫痫发作症状,其作用机制可能与上调miR-22-3p表达有关。
