IL-25在小鼠哮喘中通过Nuocyte细胞诱导Th2炎性细胞因子的表达

摘要
目的研究哮喘气道炎性因子IL-25通过nuocyte细胞对Th2炎性细胞因子的影响。方法建立小鼠哮喘模型,分离并培养nuocyte细胞,将nuocyte细胞分为PBS对照组、IL-25处理组、抗IL-25组,体外给予IL-25和抗IL-25处理。ELISA方法检测细胞上清液IL-5和IL-13的含量;Western blot检测细胞IL-5和IL-13蛋白表达;RT-PCR检测IL-5和IL-13 mRNA表达;流式细胞计数测定nuocyte细胞数量。结果 IL-25处理组nuocyte细胞IL-5及IL-13在蛋白和基因水平的表达均比对照组和抗IL-25组IL-5及IL-13表达水平高(P<0.05);IL-25处理组nuocyte细胞数量高于对照组及抗IL-25组(P<0.05)。结论 IL-25在小鼠哮喘中可通过nuocyte细胞诱导IL-5和IL-13的表达,促进哮喘的发生和发展。
