LAMB3的理化性质及其分子结构的分析

摘要
层黏连蛋白亚基β3(laminin subunit beta 3, LAMB3)在结肠癌与甲状腺癌等肿瘤发生发展中参与调控肿瘤的增殖、迁移和侵袭。本文对LAMB3的理化性质和分子结构进行了分析,为进一步研究LAMB3的功能提供了新的思路。利用ExPASy、TMHMM、SignalP、PSORTⅡ、SOPMA、SWISS MODEL等方法对LAMB3的理化性质、跨膜区、信号肽、亚细胞定位、二级结构和高级结构进行了分析。采用STRING、GEPIA分别对蛋白互作网络、肿瘤与正常组织中表达水平等进行分析。分析得出,LAMB3的分子式为C5497H8802N1702O1735S96,相对分子质量为129 572.48,理论等电点PI=7.14,不稳定指数为57.85;脂溶指数为68.06,总平均亲水性为–0.513;LAMB3是经典的分泌型蛋白,具有信号肽结构;LAMB3的亚细胞定位主要在细胞核内(60.9%);LAMB3的二级结构主要由不规则卷曲(45.82%)和α-螺旋(45.05%)组成;LAMB3包含有15个糖基化位点(12个O-糖基化位点+3个N-糖基化位点)和46个磷酸化位点;LAMB3的互作蛋白主要包括ITGB4、LAMC2、LAMA3等20个蛋白;与正常组织相比,LAMB3主要在头颈部鳞状细胞癌和胰腺癌中高表达。以上结果提示,LAMB3是一种不稳定的核内亲水碱性蛋白,具有多个修饰位点,主要与整合素家族及层黏连蛋白家族相互作用,可能参与细胞外基质受体相互作用、黏着斑、PI3K-AKT等信号通路。这将为进一步探讨LAMB3在肿瘤等疾病中的功能及作用机制提供参考。
