miR-132/212对肺癌裸鼠移植瘤生长的影响

摘要
目的探讨微小RNA-132/212(microRNA-132/212,miR-132/212)对人肺癌H1299细胞裸鼠皮下移植瘤生长的影响及可能机制。方法建立人肺癌裸鼠皮下移植瘤模型。成瘤后的裸鼠随机数字表法分为阴性对照组、空载体组和miR-132/212组,并瘤内注射相应质粒,5d干预1次,共干预4次,干预过程中3d测量1次肿瘤体积。免疫组化检测各组Ki-67、p21和Cyclin D1的表达,并计算微血管密度(microvessel density,MVD)。结果取材时,阴性对照组肿瘤体积为(1 403.33±456.19)mm3,空载体组为(1 540.24±392.35)mm3,miR-132/212组为(779.29±276.45)mm3。miR-132/212组肿瘤生长缓慢,肿瘤体积明显小于阴性对照组(F=4.503,P=0.046)和空载体组(P=0.020);空载体组与阴性对照组的肿瘤体积差异无统计学意义,P=0.625。阴性对照组增殖指数为(58.89±8.60)%,空载体组为(60.36±13.09)%,miR-132/212组为(27.89±5.63)%。miR-132/212组肿瘤细胞增殖指数显著低于阴性对照组和空载体组,F=14.584,P=0.001。阴性对照组和空载体组p21的表达显著低于miR-132/212组,P<0.05。阴性对照组和空载体组Cyclin D1的MVD显著高于miR-132/212组,P值均<0.05。结论 miR-132/212明显抑制了人肺癌H1299裸鼠皮下移植瘤生长,可能是通过上调p21的表达,下调Cyclin D1的表达,进而抑制肿瘤组织增殖和新生血管生成,抑制肿瘤生长。
