唑来膦酸对MC3T3-E1细胞增殖及Runx2、Osterix表达变化的影响

摘要
目的研究唑来膦酸(ZA)对前成骨细胞(MC3T3-E1细胞)增殖及Runx2、Osterix表达的影响。方法以不同浓度(10-3mol/L、10-8mol/L)ZA处理体外培养的MC3T3-E1细胞,采用四唑蓝比色试验(MTT)检测ZA对MC3T3-E1细胞增殖的影响;以qRT-PCR法检测成骨细胞分化相关基因Runx2、Osterix mRNA表达情况;以Western免疫印迹法检测R unx2、Osterix蛋白表达水平。结果高浓度(10-3 mol/L)ZA能抑制MC3T3-E1细胞增殖,作用48 h后与对照组具有显著性差异(P<0.05),低浓度(10-8mol/L)ZA对MC3T3-E1细胞增殖无明显影响(P>0.05)。高浓度(10-3 mol/L)ZA可降低R unx2、Osterix mR NA表达水平,与对照组相比差异具有显著性(R unx2:P<0.05;Osterix:P<0.01),而低浓度(10-8mol/L)ZA可使R unx2、Osterix mR NA表达水平升高,与对照组相比差异具有显著性(R unx2:P<0.01;Osterix:P<0.01)。结论高浓度ZA明显抑制MC3T3-E1细胞增殖,通过影响Runx2及其下游基因Osterix表达促进成骨细胞分化。
