microRNA-29a-3p通过p70s6k/S6信号通路促进乳腺癌细胞增殖

目的研究microRNA-29a-3p在乳腺癌细胞系中的表达,探讨miR-29a-3p对p70s6k/S6信号通路以及对乳腺癌细胞增殖能力的影响。方法首先通过实时聚合酶链反应检测乳腺癌细胞系MCF-7、MDA-MB-231和正常乳腺细胞MCF-10A中miR-29a-3p的表达水平,选取miR-29a-3p表达较高的乳腺癌细胞MCF-7进行后续实验;通过Western blot检测转染后p70s6k/S6信号通路中关键基因的表达,并通过MTT实验明确miR-29a-3p对MCF-7细胞增殖能力的影响。结果 miR-29a-3p在乳腺癌细胞中高表达;在MCF-7细胞中转染miR-29a-3p mimic后,p70s6k和S6的磷酸化水平显著增加,p70s6k和S6的总蛋白量无明显变化,而转染miR-29a-3p inhibitor后,呈现相反结果;此外,MTT实验显示miR-29a-3p的表达与乳腺癌细胞的增殖能力呈正相关。结论 miR-29a-3p通过p70s6k/S6信号通路促进乳腺癌细胞的增殖。
