摘要：

目的构建并鉴定pcDNA3.1-Stat3真核表达质粒,探讨信号转导和转录激活因子Stat3对Foxp3表达的影响。方法以EL4细胞cDNA为模板,PCR法获取目的基因Stat3。构建含有目的基因的T载体pMD19-T-Stat3。用HindⅢ和ECOR I双酶切pcDNA3.1载体及pMD19-T-Stat3,纯化后在T4 DNA连接酶作用下连接,构建pcDNA3.1-Stat3真核表达质粒,并进行PCR、双酶切及测序鉴定。将构建正确的pcDNA3.1-Stat3电转入EL4细胞,采用Western blot法及细胞免疫荧光法检测Foxp3的表达。结果 PCR和双酶切鉴定重组质粒pcDNA3.1-Stat3构建成功,测序鉴定插入片段无突变,序列完全正确。pcDNA3.1-Stat3转入EL4细胞后,能上调目的蛋白Stat3的表达,且能进一步促进Foxp3表达。结论 pcDNA3.1-Stat3重组质粒转化入EL4细胞后能高表达Stat3,且能诱导Foxp3的表达。
