Inhibitory mechanism of chlorogenic acid on α-glucosidase and evaluation of its glucose consumption in HepG2 cells

摘要
There is an increasing demand for plant-derived antidiabetic drugs. This study systematically evaluated the potential of chlorogenic acid as an alpha-glucosidase inhibitor and its influence on glucose metabolism in liver HepG2 cells. Kinetic analysis revealed that chlorogenic acid suppressed alpha-glucosidase activity through the mixed-type mode and monophasic process. Fluorescence quenching and circular dichroism data demonstrated that the alpha-glucosidase-chlorogenic acid complex was formed. As revealed by molecular docking, chlorogenic acid showed main interactions with amino acid residues near the alpha-glucosidase active site, mainly including hydrogen bonding and hydrophobic interactions. Chlorogenic acid also demonstrated favorable glucose consumption in HepG2 cells with minimal cytotoxicity. Our findings validate its potential as an antidiabetic nutraceutical.
