In vitro callogenesis in epicotyl segments from mahogany (Swietenia macrophylla King) plantlets using 6-benzylaminopurine and α- naphthaleneacetic acid

The objective of this work was to develop techniques for in vitro regeneration of mahogany (Swietenia macrophylla), through epicotyl segments from mahogany plantlets germinated in culture medium. The epicotyl segments were inoculated on MS medium supplemented with different combinations of BAP (6-benzylaminopurine) and NAA (α-naphthaleneacetic acid) concentrations and after 40 days of inoculation callus formation was observed at the ends of the segments. On treatments T07 (0,50 mg.L-1 NAA and 0,50 mg.L -1 BAP), T10 (0,25 mg.L-1 NAA and 1,00 mg.L-1 BAP) and T11 (0,50 mg.L-1 NAA and 1,00 mg.L-1 BAP), 100% of explants showed callus varying the intensity and texture. The callus texture from the other treatments varied from greenish on the extremes to semi compact and beige. Related to callus intensity, it was shown a variation between little and moderated, and the explants with callus varied according to the growth regulator combination (BAP and NAA), with high formation on treatments T10 (0,25 mg.L-1 NAA and 1,00 mg.L-1 BAP) and T16 (1,00 mg.L -1 NAA and 2,00 mg.L-1BAP).
