Studies of human serum albumin adsorption on cibacron blue F3GA functional microspheres

Polystyrene grafted poly (vinyl acetate) (PSt-g-PVAc) microspheres were obtained by dispersion copolymerization of PVAc macromonomer with styrene. The diameter of PSt-g-PVAc microspheres was controlled in range of 500-700 nm. Polystyrene grafted poly (vinyl alcohol) (PSt-g-PVA) microspheres were derived from alkaline hydrolysis of PVAc chains on surfaces of PSt-g-PVAc microspheres in the presence of NaOH. Dispersion stability of the PSt-g-PVA microspheres was increased in water. The CB functional microspheres were prepared via the nucleophilic reaction between PSt-g-PVA microspheres with Cibacron Blue F3GA (CB). The biggest amount of CB on surfaces of the functional microspheres was 139.22μmol/g based on the analysis of an elemental analysis instrument. The effects of origination concentration of human serum albumin (HSA), adsorption time and pH on adsorption amount of the CB functional microspheres were studied. The biggest absorption amount of CB functional microspheres was 40.9 mg/g for HSA under conditions of HSA concentration is 1.0 mg/ml and pH = 5.14. The interaction mechanism of CB functional microspheres between HSA was further discussed. Desorption ratio of 92.11% was achieved by using an eluent of 0.5 mol/L NaSCN (pH = 8.0). The CB functional PSt-g-PVA microspheres could be reused without significant decreases in the adsorption capacities.
