To establish an experimental method for the separation and detection of isoflavone compounds puerarin daidzin and daidzein in pueraria lobata root by pressurized capillary electrochromatography pCEC . Using a C18 capillary chromatographic column with NaH2 PO4 aqueous solution and methanol as mobile phases the chromatographic conditions such as mobile phase ratio mobile phase flow rate NaH2 PO4 aqueous solution concentration and pH and separation voltage were optimized. The results showed that at the mobile phase of 17. 5 mmol / L NaH2 PO4 aqueous solution pH 4. 0 methanol = 55 45 V / V separation voltage of 3 kV mobile phase flow rate of 8 μL / min detection wavelength of 250 nm puerarin daidzin and daidzein had a good linear relationship in 200-1000 μg / mL with the correlation coefficient r of 0. 9960-0. 9982 and the average recovery rates of 98. 6%-100. 9% respectively. RSDs were 3. 1%-3. 5% . The method is suitable for the separation and detection of isoflavones in pueraria lobata separation and analysis of isoflavones in pCEC in the future. © 2021, Youke Publishing Co.,Ltd.
