Extraction of Volatile Oil from Podocarpus macrophyllus by Microcapsule Aqueous Two-phase System and Its Composition Analysis

Total volatile oil from the branches and leaves of Podocarpus macrophyllus was extracted by the combination of microcapsule technology and aqueous two-phase extraction technology. After screening, β-cyclodextrin and sodium sulfate was selected as extraction agents in aqueous two-phase system. On the basis of single factor experiment, the extraction conditions for volatile oil from Podocarpus macrophyllus were optimized by four-factor and three-level response surface methodology. The optimum extraction conditions were obtained as follows: extraction time 36 min, mass fraction of β-cyclodextrin 42%, mass fraction of sodium sulfate 15%, and extraction temperature 33℃. Under these conditions, the yield of total volatile oil from Podocarpus macrophyllus was 1.369%. A total of 22 compounds were identified by GC-MS, among which, containing 18 kinds of terpenes and their oxygen-containing derivatives and 4 kinds of aliphatic compounds.
