Screening and Identification of Poly Malic Acid Producing Strains

Poly malic acid (PMA) producing strains were isolated and screened from the campus. Twenty-one PMA producing strains were obtained by centrifugal tube primary screening, and it was preliminarily judged that the strains I-2, Ⅰ-13a and Ⅲ-16 had strong PMA producing capacity. It was determined that strain Ⅰ-13a had the strongest PMA production capacity, and the yield reached (43.08±0.36) g/L after shake flask secondary screening. The high yield strains were identified by morphological observation and molecular biological identification. The results showed that the strain was light pink at the initial stage of growth, and turned green and dark at the later stage. The cells were budding, oval and single. The length of ITS nucleotide sequence was 560 bp, and the homology similarity with Aureobasidium melanogenum was 97%. Finally, the strain was identified as Aureobasidium melanogenum and named as ZUST-HD.
