SVF-gel促进糖尿病鼠创面愈合的初步实验研究

摘要
目的探讨SVF-gel治疗糖尿病大鼠慢性创面的效果及机制。方法 36只雄性SD大鼠,采用一次性腹腔注射链脲佐菌素(STZ)55 mg/Kg制备糖尿病模型,1周后于糖尿病鼠背部两侧各制备1个直径为1.2 cm的圆形全层皮肤缺损创面。随机分为3组,每组12只,分别采用SVF-gel(A组)、SVF悬液(B组)、生理盐水(C组)处理创面。观察各组创面愈合情况并计算创面愈合率,采用HE染色、免疫组织化学染色观察创面内肉芽组织形态学改变和新生血管情况。结果 A、B组各时间点创面愈合率均优于C组(P<0.05),A组各时间点创面愈合率均优于B组(P<0.05)。相比C组,A、B组炎症细胞浸润较少,再上皮化程度明显,新生血管明显增多,肉芽组织结构致密,且A组组织学改变最为明显。结论 SVF-gel治疗可以通过减轻炎症反应,促进创面内血管生成,增加肉芽组织形成来加速慢性创面的愈合。
