摘要
The introduction of fluorine to the drug molecules endows the compounds with better metabolic stability and bioavailability. However, naturally occurring fluorides are rare and there are challenges in identification of natural fluorination pathways and their corresponding fluorinases. Synthetic biology strategies have been applied to construct artificial fluorination pathways via incorporation of fluorine into the polyketide backbone, which provides new routes to synthesize fluorides with complex structures. This review summarizes the latest research progresses in mining natural fluorination pathways and the engineering of artificial fluorination pathways.
