APPLICATION OF THE FLUORESCENT-PROBES DSM AND DSP-6 FOR STUDIES OF THE STRUCTURAL AND FUNCTIONAL-CHANGES OF THE ERYTHROCYTE-MEMBRANE

The affinity of erythrocyte beta-adrenoreceptors. to beta-blocking agents was enhanced and the parameters of their binding by a fluorescent probe were investigated. The data obtained suggest the possibility of using erythrocytes for in vivo estimation of the functional state of the adrenoactive system in man and animals. A detailed characterization of the spectral properties of DSM and DSP-6 in isotonic phosphate buffer (pH 7.0) and in erythrocyte suspension was carried out. The maximum of the emission spectrum of the probes in buffer was observed at 610 nm (lambda(ex) 450 nm). The emission spectrum of the probes in erythrocyte suspension was shown to consist of two bands with the maxima at 560 nm (lambda(ex) 470 nm) and 620 nm (lambda(ex) 459 nm). Optimal conditions for the application of fluorescent probe in erythrocyte membrane studies were determined. The binding of a probe to the erythrocyte membrane was studied at high and low adrenoactivity in guinea pig and rats; in pathological states (ischemic heart disease, neurocirculatory distony etc); in extreme conditions (antiorthostatic hypokinesia etc). The differences in the parameters of binding of the probe to erythrocytes were found in observed groups of animals, patients and volunteers as compared with healthy subjects. The obtained results indicate structural-functional changes in membrane, including the state of beta-adrenoreceptors. It is concluded that DSM and DSP-6 probes are promising in studies of the cellular membrane and functional state of beta-adrenoactive system in man and animals.
