摘要
A copper(II)-alpha,beta,delta,gamma-tetrakis (4-sulfonatophenyl) porphine (TPPS) complex had an absorption maximum wavelength of the Soret band at 113 nm (molar absorptivity: 4.76 X 10(5) mol(-1) dm(3) cm(-1)). Also, in the presence of L-ascorbic acid as a reaction accelerator, the copper(II) ion quantitatively formed a stable complex with TPPS within one minute at room temperature. Based on these results, the spectrophotometric detection-FIA system of copper(II) determination in serum was developed, The proposed FIA is a double-line manifold system consisting of a line for spectral shift and one for complex formation. Recommended procedure: A 0.2 cm(3) serum sample is placed in a beaker. A 0.1 cm(3) aliquot of 1 mol dm(-3) hydrochloric acid aqueous solution is added and then mixed. After standing for 5 min at room temperature, 0.1 cm(3) of a 1.2 mol dm(-3) trichloroacetic acid aqueous solution is added to this mixture and centrifuged (2000 rpm, 5 min). After filtering, 0.1 cm(3) of 1 mol dm(-3) ammonium acetic acid buffer solution is then added to 0.1 cm(3) of the filtrate. Fifty mu l of the mixture solution is injected into the FIA system. At least eighteen samples can be determined within one hour {the RSD: 1.6% (5 determinations). This method was applied to dog and human serum with satisfactory results.
