DETERMINATION OF VARIOUS NEUROPLEGICS AND THE BETA-BLOCKER CARAZOLOL FROM PIGS KIDNEYS BY MEANS OF HPLC

To detect and quantify various neuroplegics and the beta-blocker Carazolol, we first of all extracted pig's kidneys with sulfuric aqueous acetonitrile. After degreasing with n-hexane, the extract was turned alcaline, extracted with tertiary butyl-methyl ether, evaporated to dryness, dissolved the residue in methanol and submitted to centrifugation after having added phosphoric acid/acetonitrile (1:1). The target substances are determined from the lapping solution through HPLC (UV detection). Fortification experiments (10-83 mug/kg for the neuroplegics, 2-15 mug/kg for Carazolol) yielded recoveries between 76 and 96%. Detection limits as defined and deduced from DIN 32645 were 3 to 7 mug/kg for the neuroplegics and 0.5 mug/kg for Carazolol. The limits of Determination as per DFG (1991) amounted to between 9 and 18 mug/kg for the neuroplegics, and to 1.5 mug/kg for Carazolol. One laboratory employee can manage the complete processing of 4 samples each work day.
