A test kit for the quantitative determination of potato leafroll virus (PLRV) is described. The reliability of the test is characterized by the precision between the series, using the variation of the mean value (E405nm = 0.00 to 0.12) for five dilution steps of the reference solution. Various methods can be used to analyze the test data for differentiation of three potato genotypes with artificial PLRV infection. After logarithmic transformation of the extinction values and computation by variance analysis, for about eight weeks after infection cv. Astilla is reliably distinguished from cvs. Adretta and Galina. The relative virus concentration and, hence, the quantitative resistance of cvs. Adretta and Galina differ only in five out of nine samples. Methods according to which the extinction values are converted into concentrations by means of the standard curve do not give better chances for differentiation.
