Seedlings of P. vulgaris cv. Canadian Wonder were incubated with 4-14C-estrone, 4-14C-estradiol-17.beta. and 6,7-3H-estrone sulphate. Evidence from TLC, column chromatography on Lipidex, radio-GLC and crystallization to constant specific activity suggests that estrone and estradiol-17.beta. are interconvertible in plant tissues. A tissue homogenate supplemented with cofactors evidently can metabolize estrone. Analysis by gas chromatography and combined gas chromatography-mass spectrometry demonstrated unequivocally that the major product was estradiol. These data represent the 1st evidence from full mass spectrometry that estrogens can be metabolized by plant tissues.
