CELLULOSE DIGESTION BY AN EXTREMELY THERMOPHILIC ANAEROBIC BACTERIUM

Cellulase production, insoluble cellulose digestion and growth of an extremely thermophilic cellulolytic anaerobe, strain NA10 (optimum temperature, 73-degrees-C), were studied in continuous cultures at different hydraulic retention times, keeping the influent cellulose concentration constant at 10 g/l. Most of cellulase produced by strain NA 10 attached to cell mass and cellulose. Cellulase activity in the culture broth was 0.07-0.1 unit/ml, while cellulase productivity increased with an increase in retention time until nearly the washout point with a maximum productivity of 8.1 unit/l.h. The yield of acetic acid, the main product of cellulose digestion, was 32-40% based on the carbon balance. Data of continuous culture in the steady state were analyzed and the following kinetic parameters for cellulose digestion by strain NA10 were obtained: Maximum specific growth rate=0.13 h-1, maximum specific cellulose consumption rate=2.0 mg glucose equivalent/mg cell.h, saturation constant=200mg cellulose/l, true growth yield=0.08mg cell/mg glucose equivalent and maintenance coefficient = 0.46 mg glucose equivalent/mg cell.h.
